Effects of trace element deficiency on drug-metabolizing enzymes in rats

We investigated the effects of trace element deficiency on drug-metabolizing enzymes in rats. Weanling rats were fed a selenium-deficient diet (Torula yeast) or a normal diet (0.5μg selenium) until 20 weeks. After six weeks of the selenium-deficient diet, an increase in aniline hydroxylase activity was observed in the selenium-deficient rats, but the activity of aminopyrine N-demethylase was reduced. In three-week-old Wistar rats randomly assigned to three groups: a zinc-adequate ad libitum group, a zinc-adequate pair-fed group, and a zinc-deficient group, zinc deficiency resulted in decreased aminopyrine N-demethylase, aniline hydroxylase and p-nitroanisole o-demethylase activities. Moreover, Western immunoblot analysis using CYP 2C11 antibody revealed an inhibition of CYP 2C11 protein synthesis in zinc-deficient rats at five weeks. The results of this study suggest that drug-metabolizing enzymes are affected by selenium and zinc deficiency in rats.
